you can't configure it in tuxguitar, since it isn't who produce the sounds, but at the MIDI synthesizer..
In tuxguitar, at menu Tools -> Settings, in the Sound section there is a "MIDI Port" select field. the MIDI Port selected there is who do the job.
These available MIDI Ports could change depending on what tuxguitar's plugins do you have installed, on what OS are you running, and what software/hardware do you have installed at your OS.
So the answer there is yes in some MIDI Port cases, or no in others..
The question is, what operating system do you have ?
Anyway, i don't think you can directly add MP3 format. i think you would need to convert them to WAVE or any other raw format, and add them into a Soundfont (*.sf2), Downloadable Sound (*.dls ), etc..
